My advice would be, read the profile of the person you like. Check for similar interests. And start talking about them.

When you view a profile of interest, try sending a message only to those that have visited Mingle2 recently, if at the top of their profile it says - "Last seen over a month ago" - I would give those a miss because they could be dating someone already, or having a break, and that's why you are not receiving any replies.
